Microfluidic Sperm Sorting is a method designed to select functional sperm with low DNA fragmentation by using a specific chip. It enables to retrieve the sperm of the highest quality, thereby increasing the chances of successful infertility treatment.
The method is based on the principle of natural sperm selection through passing across microbarriers (micropores, microchannels) simulating the natural environment of the female reproductive system (fallopian tubes). Sorted sperm have better morphology, genetic quality, and functional properties compared to unsorted sperm.
Microfluidic Sperm Sorting is also a gentle alternative to standard sperm processing, as it does not require any preliminary examination of the semen sample or the use of devices that may increase oxidative stress in sperm, which could negatively affect their quality.
The chip has an inlet for the sample, which is connected to an output through a microfluidic channel or membrane with micropores.
The untreated semen sample
is pipetted into the inlet, from which the present sperm must actively move through the mentioned microbarriers to the end of the tunnel
with fertilisation medium.
Healthy, motile sperm is sorted out into the output from low-quality sperm, which stays trapped in the channel or beneath the membrane.
The method effectively reduces the proportion of sperm with fragmented DNA and, in indicated cases, significantly increases the proportion of euploid embryos (embryos with the correct number of chromosomes) and the overall success rate of the cycle.
Permanent embryo monitoring is used for continuous tracking of embryo development from the moment of fertilization until stages suitable for transfer into the woman's uterus.
Permanent embryo monitoring is performed by using a camera integrated into the incubator, which captures the development of embryos and enables a better detection of embryos with a non-standard development and lower chances for successful implantation, which cannot be recognized during conventional microscopy.
At GENNET clinic, we use the unique Embryoscope+ incubator for permanent embryo monitoring, equipped with a neural network for embryo selection - IDAScore software.
In the Embryoscope+ incubator, the embryos are individually cultured in chambers with a built-in camera. The system also continuously monitors the cultivation conditions of embryos – temperature, CO2, O2 – providing an optimal environment for the embryo development. Due to its unique technology, embryologists have access to a complete record of embryo development without disrupting the optimal conditions for their growth.
During cultivation, the IDAScore software assigns each embryo a rating based on a deep learning algorithm to predict the viability of embryos cultured in the Embryoscope+ incubator.
IDAScore identifies embryos with the highest chance of implantation, serving as a complement to clinical decision-making and the final embryologists’ assessment. IDAScore objectively "compares" the given embryo with other embryos that have similar developmental patterns, generating a score correlating with the probability of implantation. The outputs generated by Embryoscope+ are excellent tools for embryologists to select the most suitable embryo for transfer.
